- From Orlando: Exit #68 (formerly exit 27) from I-4 West at Lake Buena Vista. Turn left to S.R. 535, stay in left lane, turn left at light onto Vineland Avenue (at Little Lake Bryan), just before the Holiday Inn.
- From Tampa: Exit #68 (fromerly exit 27) from I-4 East at Lake Buena Vista. Drive straight to Shrine (no turns).
- Coming north on S.R. 535 from S.R. 192: Vineland Avenue is the last road before I-4. Just past the Holiday Inn, turn right at the traffic light onto Vineland Avenue (at Little Lake Bryan).
- From Lake Buena Vista Hotels: Take S.R. 535 south (towards I-4), go under I-4 and continue in the left lane to Vineland Avenue (about 300 yards). Turn left at the traffic light onto Vineland Avenue (at Little Lake Bryan) and proceed to the Shrine entrance.
- From the International Drive Area & Southwest Area: Follow International Drive south to Little Lake Bryan Parkway (1.1 miles south of the Mission Club). Turn right, then right again and follow the Parkway to the Shrine.
